目的:本研究以喉癌细胞系Hep2为研究对象,通过体外药物敏感实验分析化疗疗效的时间依赖性和剂量依赖性特点,检测3种化疗药物对喉癌细胞的杀伤作用。方法:用CCK8试剂盒体外药物敏感实验,测定5FU、紫杉醇、卡铂在不同浓度梯度和接触时间下药物的抑制率,选择最佳给药方式。结果:体外药敏实验结果显示:5FU对细胞的杀伤是受剂量和时间依赖性,而紫杉醇主要受接触时间的影响,卡铂主要受药物剂量的影响。结论:药物浓度和接触时间对药物敏感性的结果均有影响,体外药物敏感实验可为临床用药提供指导。
Objective: In this study, the laryngeal carcinoma cell line Hep2 was used as the research object, and the chemosensitivity test in vitro was used to analyze the time-dependent and dose-dependent effects of chemotherapy. Methods: In vitro drug sensitivity test of CCK8 kit was used to determine the inhibition rate of 5FU, paclitaxel and carboplatin under different concentration gradients and contact time, and the optimal dosage was selected. Results: In vitro susceptibility test results showed that 5FU killed cells in a dose and time-dependent manner, while paclitaxel was mainly affected by exposure time. Carboplatin was mainly affected by drug dose. Conclusion: The drug concentration and the contact time have an impact on the results of drug sensitivity. In vitro drug sensitivity test can provide guidance for clinical use.
