目的 探讨microRNA-433对人结肠癌细胞Sw620生物学行为的影响.方法 应用细胞转染技术,使Sw620细胞中高表达miR-433并通过实时PCR检测转染效率.将细胞分为con组和高表达miR-433的433组.采用MTT法检测细胞增殖,流式细胞仪检测细胞凋亡及细胞周期,Transwell小室实验检测细胞迁移.结果 Sw620细胞转染48h后,与con组相比,433组中miR-433表达升高(P< 0.001),细胞增殖受到抑制(P<0.05),细胞迁移受到抑制(P<0.01),细胞凋亡及细胞周期无明显变化(P> 0.05).结论 miR-433抑制Sw620细胞的增殖和迁移,但对细胞凋亡及细胞周期无明显作用.“,”Objective To study the effect of microRNA-433 on biologic behaviors of human colon cancer cell line Sw620.Methods The cell transfection assay was used to make the miR-433 mimic into Sw620 and the transfection effect was detected by real-time quantitative PCR.The cells were divided into two groups,control group and miR-433 group highly expressed miR-433.MTF assay was used to evaluate the proliferation.Flow cytometry was used to detect the cell apoptosis and cell-cycle distribution and the transwell chamber assay was used to investigate the migration ability.Results 48 hours after transfection of miR-433,the expression level of miR-433 in Sw620 cells was increased obviously in miR-433 group,and the growth rate and migration rate of the cells were inhibited(P < 0.05),but with no obvious changes for the cell apoptosis rate and the cell-cycle distribution(P > 0.05).Conclusion miR-433 inhibited the proliferation and migration of Sw620 cells but not for cell-cycle distribution and cell apoptosis.
