目的:建立一种简单、快捷、灵敏的人血浆中尼莫地平的LC/MS测定方法。方法:血浆样品用乙醚进行萃取浓缩处理;色谱分离采用ZorbaxSB-C18柱,流动相为甲醇-水(75:25),流速为0.8mL.min-1;分离物检测采用单四级杆质谱仪,使用大气压化学电离源,负离子模式,选择离子检测(SIM)方式定量分析,监测离子的质荷比(m/z)为417.1(尼莫地平)和359.1(尼群地平,内标)。结果:血浆中尼莫地平测定的线性范围为0.25~50μg.L-1,定量限为0.25μg.L-1,精密度和准确度满足生物样品分析要求。结论:所建方法简单、灵敏、专属性好,适用于尼莫地平的药动学研究。
Objective: To establish a simple, rapid and sensitive method for the determination of nimodipine in human plasma by LC / MS. Methods: The plasma samples were extracted and concentrated by ether. The chromatographic separation was carried out on a Zorbax SB-C18 column with a mobile phase of methanol-water (75:25) at a flow rate of 0.8 mL.min-1. The single quadrupole mass spectrometer (M / z) of 417.1 (nimodipine) and 359.1 (nitrendipine, internal standard) were monitored using atmospheric pressure chemical ionization source, negative ion mode and selective ion detection (SIM) Results: The linear range of nimodipine was 0.25 ~ 50μg.L-1 and the limit of quantification was 0.25μg.L-1. The precision and accuracy of the assay met the requirements of biological sample analysis. Conclusion: The method is simple, sensitive and specific, and is suitable for pharmacokinetic studies of nimodipine.
