目的:观察涤痰化瘀汤含药血清对人食管癌Eca-109细胞生长的抑制作用及对ras p21、突变型p53基因表达的影响。方法:以人食管癌Eca-109细胞为材料,采用MTT法,观察涤痰化瘀汤含药血清对细胞生长的抑制率;应用流式细胞仪检测涤痰化瘀汤对人食管癌Eca-109细胞ras p21、突变型p53基因表达的影响。此外以ICR纯种小鼠的骨髓细胞为材料,以微核为指标,观察涤痰化瘀汤的致变作用,以进行药物安全性评估。结果:涤痰化瘀汤含药血清对人食管癌Eca-109细胞有抑制作用;含药血清组与对照血清组比ras p21、突变型p53基因表达均降低;涤痰化瘀汤药物组的微核率与正常对照组比无显著差别。结论:涤痰化瘀汤含药血清对人食管癌Eca-109细胞生长有明显的抑制作用且对ras p21、突变型p53基因表达有影响。涤痰化瘀汤无致变作用。
OBJECTIVE: To observe the inhibitory effect of serum from Ditan Huayu Tang on the growth of human esophageal carcinoma Eca-109 cells and its effect on the expression of ras p21 and mutant p53 gene. Methods: Human esophageal cancer Eca-109 cells were used as materials, MTT assay was used to observe the inhibitory rate of Ditan Huayu Decoction-containing serum on cell growth. Using flow cytometry to detect the expression of Eca- 109 cells ras p21, mutant p53 gene expression. In addition, the bone marrow cells of ICR purebred mice were used as materials, and micronuclei as indexes to observe the mutagenic effect of Ditan Huayu Decoction for drug safety evaluation. Results: The serum of Ditan Huayu decoction could inhibit the proliferation of human esophageal cancer cell line Eca-109; the expression of ras p21 and mutant p53 gene in serum of the drug-containing serum group was lower than that of the control serum group; There was no significant difference between micronucleus rate and normal control group. CONCLUSION: The serum of Ditan Huayu Decoction can significantly inhibit the growth of human esophageal carcinoma cell line Eca-109 and affect the expression of ras p21 and mutant p53 gene. Ditan decoction soup no change effect.
