目的了解大剂量吡哆醇(PN)对雄性大鼠王殖系统的作用。方法选用成年雄性SD大鼠40只,随机分为4组,分别经腹腔注射0,100,300,600mp.kg-1.d-1的盐酸吡哆醇。结果给药15d,300mp和600mg组的睾丸、附睾、精囊和前列腺减重,至给药30d,各实验组除上述改变外,600mp组的星丸体积缩小;光镜下,100mg组无明显异常改变,随着剂量和给药时间的增加,精曲小管由精子细胞脱落发展到精子细胞、精母细胞变性坏死,电镜下见精子释放延迟和异形精子,Sertoli细胞肿胀、微管稀少,内质网异位和扩张,结论PN可能通过破坏细胞骨架而损伤Sertoli细胞,抑制星酮作用及直接损伤生精细胞而引起雄性大鼠生殖毒性。
Objective To investigate the effect of high dose pyridoxine (PN) on the king rat reproductive system in male rats. Methods Forty adult male Sprague-Dawley rats were randomly divided into 4 groups, which were injected intraperitoneally with 0, 100, 300 and 600 m. kg-1. d-1 pyridoxine hydrochloride. Results The weight of the testis, epididymis, seminal vesicles and prostate was decreased in the 15th, 300th and 600th mg groups, and the volume of the astrocytes in the 600mp group was reduced to the level of 30 days after administration. No significant changes were found in the 100mg group , With the dose and dosing time increases, the seminiferous tubules from the sperm cells off to the development of sperm cells, spermatocytes degeneration and necrosis, delayed sperm delivery and abnormal shaped sperm under electron microscopy, Sertoli cell swelling, microtubules, endoplasmic reticulum Ectopic and dilatation. Conclusions PN may damage Sertoli cells by disrupting the cytoskeleton, inhibit the action of star-ketone and directly injure spermatogenic cells, which may cause reproductive toxicity in male rats.
