乙型肝炎病毒(HBV)的长期携带者可发生肝细胞癌(HCC)。曾有报道在HCC中HBV的复制和病毒核心(C)基因表达显著低下。作者曾发现肝癌细胞中HBV增强子(EN)的活性低下,因此认为HCC中HBV复制和基因表达低下可能是由于HBV EN活性减弱所致。鉴于腺病毒ElA蛋白具有转化细胞作用,并可调控多种细胞及病毒基因的表达,本文研究了ElA蛋白对EN2的作用。首先构建了一系列含有编码ElA全部结构区域的表达载体pElASV以及有HBVEN2片段插入pTHCAT质粒中CAT基因下游的质粒pTKCATC_1。两质粒共转染人肝癌细胞Huh6后,利用检测CAT活性观察
Long-term carriers of Hepatitis B virus (HBV) can develop hepatocellular carcinoma (HCC). It has been reported that HBV replication and viral core (C) gene expression are significantly lower in HCC. The authors have found that HBV enhancer (EN) activity in hepatocellular carcinoma cells is low, so that low HBV replication and gene expression in HCC may be due to weakened HBV EN activity. In view of adenovirus ElA protein has a role in transforming cells, and can regulate a variety of cell and viral gene expression, the paper studied the effect of ElA protein on EN2.
