探讨脂肪酸对人卵巢颗粒细胞生长的影响。不同剂量的游离脂肪酸处理颗粒细胞 1~3天 ,用台盼蓝排除法对活细胞进行计数。用染色体DNA断裂和细胞形态学改变确定细胞凋亡。检测脂肪酸代谢产物酰基辅酶A是否直接引起细胞凋亡。测定与细胞凋亡相关的Bcl 2和Bax蛋白质表达水平。结果表明 ,饱和脂肪酸通过其代谢产物酰基辅酶A呈剂量依赖性的引起颗粒细胞凋亡 ,并导致Bcl 2水平下降和Bax水平升高。说明饱和脂肪酸对卵巢颗粒细胞有直接致凋亡作用。提示 ,女性肥胖和胰岛素抵抗患者出现的性功能障碍可能与饱和脂肪酸水平增高有关
To investigate the effect of fatty acids on the growth of human ovarian granulosa cells. Different doses of free fatty acid treatment of granulosa cells 1 to 3 days, with trypan blue exclusion method of living cells were counted. Apoptosis was determined by chromosomal DNA fragmentation and morphological changes. To detect whether the fatty acid metabolite acyl-CoA directly causes apoptosis. Bcl 2 and Bax protein expression levels associated with apoptosis were determined. The results showed that saturated fatty acids caused apoptosis of granulosa cells in a dose-dependent manner through its metabolite acyl-CoA, which resulted in the decrease of Bcl 2 and the increase of Bax. Saturated fatty acids on ovarian granulosa cells caused by a direct role of apoptosis. Suggesting that sexual dysfunction in women with obesity and insulin resistance may be related to increased levels of saturated fatty acids
