目的:研究Hes1蛋白在大鼠肌源性干细胞(MDSCs)体外诱导分化为神经样细胞过程中的表达差异。方法:大鼠乳鼠骨骼肌消化后体外培养增殖,第5代传代后细胞分为两组:对照组完全培养基培养,实验组加入诱导剂培养。6d后观察两组细胞的形态变化,并进行神经元特异性烯醇化酶(neuron-specific enolase,NSE)免疫细胞化学鉴定。然后通过免疫荧光和Western Blot技术观察Hes1蛋白的表达差异。结果:实验组细胞呈NSE染色阳性,对照组基本不表达;免疫荧光和Western Blot结果显示,与对照组比较,实验组Hes1蛋白表达较强(P<0.05)。结论:在体外,大鼠肌源性干细胞分化为神经样细胞过程中Hes1蛋白表达降低。
OBJECTIVE: To study the difference of expression of Hes1 protein in differentiation of rat myogenic stem cells (MDSCs) into neuron-like cells in vitro. Methods: Skeletal muscle of rat suckling mice were cultured and proliferated in vitro. The cells were divided into two groups after passage 5: the control group was cultured in complete medium and the experimental group was cultured with inducer. After 6 days, morphological changes of the two groups were observed and neuron-specific enolase (NSE) immunocytochemistry was performed. The difference of Hes1 protein expression was observed by immunofluorescence and Western Blot. Results: The cells in the experimental group were positive for NSE staining but not in the control group. The immunofluorescence and Western Blot showed that the expression of Hes1 protein in the experimental group was stronger than that in the control group (P <0.05). CONCLUSION: In vitro, the expression of Hes1 protein is decreased in rat myogenic stem cells differentiating into neuroid cells.
