目的建立高效液相色谱法测定大鼠血浆和尿液中多粘菌素B1(多肽类抗生素)的含量。方法用Onyx Monolithic C18色谱柱,Torrance C18预柱,流动相为乙腈-四氢呋喃-水(50∶25∶25),流速1.0 mL.min-1,激发波长260 nm,发射波长315 nm。结果血浆和尿液中多粘菌素B1的线性范围分别是0.12~4.00μg.mL-1(γ=0.998)和0.12~4.00μg.mL-1(γ=0.9964),日间与日内精密度均<15%。结论该方法快速、准确、稳定、特异,适用于多粘菌素B1在大鼠体内的药代动力学研究。
Objective To establish a method for the determination of polymyxin B1 (polypeptide antibiotics) in rat plasma and urine by high performance liquid chromatography. Methods Onyx Monolithic C18 column and Torrance C18 column were used. The mobile phase consisted of acetonitrile-tetrahydrofuran-water (50:25:25) with a flow rate of 1.0 mL · min-1. The excitation wavelength was 260 nm and the emission wavelength was 315 nm. Results The linear ranges of polymyxin B1 in plasma and urine were 0.12 ~ 4.00μg.mL-1 (γ = 0.998) and 0.12 ~ 4.00μg.mL-1 (γ = 0.9964) respectively. The intra- and inter- All <15%.
