目的制备6C型肺炎链球菌抗血清,并进行鉴定。方法将经美国单克隆抗体方法鉴定为6C型的肺炎链球菌制成菌体抗原免疫家兔,制备抗血清;将抗血清与6A、6B和6C型肺炎链球菌进行荚膜肿胀和凝集试验;用6A、6B及6A和6B型肺炎链球菌吸附血清后,再次检测其与各型肺炎链球菌的凝集效价。结果免疫前的兔血清与6A、6B和6C型肺炎链球菌均未出现荚膜肿胀及凝集反应。免疫后的血清与6A、6B和6C型的凝集效价分别为1∶64、1∶32和1∶64;经6A或6A和6B型吸附后,与6A、6B和6C型的凝集效价分别为(-)、(-)和1∶8;经6B型吸附后,与6A、6B和6C型的凝集效价分别为1∶16、(-)和1∶16。结论6C型为新的肺炎链球菌血清型,已成功制备出6C型抗血清,可用于血清分型研究。
Objective To prepare 6C pneumococcal antiserum and identify it. Methods Rabbit was immunized with Streptococcus pneumoniae which was identified as 6C by the method of American monoclonal antibody to prepare antiserum. The antisera was subjected to capsular swelling and agglutination with Streptococcus pneumoniae 6A, 6B and 6C. Serotypes 6A and 6B, and 6A and 6B pneumococci were used to adsorb the serum, and the agglutination titer against each type of Streptococcus pneumoniae was tested again. Results There was no capsular swelling and agglutination in rabbit serum before immunization and in Streptococcus pneumoniae 6A, 6B and 6C. The agglutinated titers of the immunized sera with 6A, 6B, and 6C were 1:64, 1:32, and 1:64, respectively; after agglutination with type 6A or 6A and 6B, the agglutination titer with type 6A, 6B and 6C (-), (-) and 1: 8, respectively. The agglutination titers with type 6A, 6B and 6C after type 6B adsorption were 1:16, (-) and 1:16, respectively. Conclusion Type 6C is a new serotype of Streptococcus pneumoniae, 6C antiserum has been successfully prepared for serotyping.
