目的:准确、快速测定盾叶薯蓣中薯蓣皂苷的含量。方法:以薯蓣皂苷含量为评价指标,采用高效液相色谱法对薯蓣皂苷的检测条件进行了优化;同时,运用3因素3水平的正交实验,对薯蓣皂苷的提取条件进行了研究。结果:优化后的检测条件为:C18色谱柱(250mm×4.6mm,5μm),柱温30℃,乙腈-水(45∶55),进样量20μL,流速1.0mL.min-1,检测波长210nm。最优提取条件为药材称取量2g,用80%乙醇超声提取40min。结论:优化后的提取及检测条件能快速准确测定盾叶薯蓣中薯蓣皂苷的含量。
Objective: To accurately and rapidly determine the content of diosgenin in Dioscorea zingiberensis. Methods: Using diosgenin as evaluation index, the detection conditions of diosgenin were optimized by high performance liquid chromatography. At the same time, the extraction conditions of diosgenin were studied by using orthogonal experiment of 3 factors and 3 levels. Results: The optimized detection conditions were: C18 column (250 mm × 4.6 mm, 5 μm), column temperature 30 ℃, acetonitrile-water (45:55), injection volume 20 μL, flow rate 1.0 mL · min- 210nm. The optimal extraction conditions for the herbs weighing 2g, 80% ethanol extraction 40min. Conclusion: The optimized extraction and detection conditions can quickly and accurately determine the content of diosgenin in Dioscorea zingiberensis.
