目的探讨化疗药物、细胞因子诱导的杀伤细胞(CIK细胞)、特异性细胞毒性T淋巴细胞(CTL)对乳腺癌MCF-7细胞CD55high亚群的杀伤效果。方法用不同血药浓度的化疗药物多西他赛、表阿霉素和氟尿嘧啶杀伤乳腺癌MCF-7细胞;抽取人外周血诱导培养CIK细胞,杀伤CD55high细胞和CD55low细胞;术中取患者腋下淋巴结体外诱导培养肿瘤特异性CTL,检测CTL对CD55high细胞和CD55low细胞体外杀伤活性,分析化疗药物、CIK细胞、特异性CTL对CD55high亚群细胞的杀伤效果。结果多西他赛、表阿霉素和氟尿嘧啶对CD55high亚群细胞杀伤率明显低于其对MCF-7细胞杀伤率;CIK细胞CD55high亚群细胞杀伤率(42.72±4.36)%;特异性CTL对CD55high、CD55low和MCF-7细胞杀伤率分别为(52.86±4.45)%、(22.41±2.83)%、(21.67±4.15)%,CD55high组明显高于CD55low和MCF-7组细胞的杀伤率(P<0.01)。特异性CTL组与CIK细胞组和多西他赛(0.4μg/mL)组3组比较,有显著性差异(P<0.01)。结论特异性CTL对CD55high细胞具有特异性杀伤效果。
Objective To investigate the cytotoxicity of chemotherapeutic drugs, cytokine-induced killer cells (CIKs) and cytotoxic T lymphocytes (CTLs) on the CD55high subsets of breast cancer MCF-7 cells. Methods MCF-7 cells were killed with docetaxel, epirubicin and fluorouracil using different concentrations of chemotherapeutic drugs. CIK cells were induced from peripheral blood to kill CD55high cells and CD55low cells. Tumor-specific CTLs were cultured and induced in vitro by lymph nodes. The cytotoxicity of CTL on CD55high cells and CD55low cells was detected. The cytotoxicity of chemotherapeutic agents, CIK cells and CTLs on CD55high subpopulations was analyzed. Results The killing rates of docetaxel, epirubicin and fluorouracil on CD55high subpopulations were significantly lower than those of MCF-7 cells. The cytotoxicity of CD55high subpopulation in CIK cells was 42.72 ± 4.36% The killing rates of CD55high, CD55low and MCF-7 cells were (52.86 ± 4.45)% and (22.41 ± 2.83)% and (21.67 ± 4.15)%, respectively. <0.01). Specific CTL group and CIK cell group and docetaxel (0.4μg / mL) group 3 group, there was a significant difference (P <0.01). Conclusion Specific CTL has a specific killing effect on CD55high cells.
