目的 研究羟基积雪草甙 (madecassoside ,Md)对人皮肤成纤维细胞 (humanskinfibrob last,HSFb)的增殖及胶原蛋白合成的影响。方法 XTT法研究Md对HSFb增殖的影响 ;放射免疫的方法测定实验组和空白对照组细胞上清液中Ⅰ型和Ⅲ型前胶原氨基端肽原 (procollagenⅠandⅢofaminoterminalpropeptide ,PINP ,PⅢNP)的含量。 结果 (1 )Md在浓度高达 2 0 0 μg/ml时仍未见任何细胞毒性。 (2 )与空白对照组相比 ,Md对HSFb有明显促进增殖作用 (P <0 0 5) ,有效刺激HSFbPINP和PⅢNP的分泌。 (3)Md刺激靶细胞PINP和PⅢNP分泌的生物学作用呈现明显的量效关系。结论 Md在刺激体外培养HSFb增殖及Ⅰ、Ⅲ型胶原合成中均有明显的生物活性
Objective To investigate the effects of madecassoside (Md) on the proliferation and collagen synthesis of human skin fibroblast (HSFb). Methods XTT method was used to study the effect of Md on the proliferation of HSFb. Radioimmunoassay was used to determine the content of procollagen I and III of amino terminal propeptide (PINP, P III NP) in the supernatants of the experimental and control groups. Results (1) Md did not show any cytotoxicity at concentrations as high as 200 μg/ml. (2) Compared with the blank control group, Md significantly promoted the proliferation of HSFb (P < 0.05), and effectively stimulated the secretion of HSFbPINP and PIIINP. (3) The biological effect of Md on the secretion of PINP and PIIINP in target cells showed a clear dose-effect relationship. Conclusion Md has obvious biological activity in stimulating the proliferation of HSFb and the synthesis of type I and type III collagen in vitro.
