目的:筛选出能明显影响肿瘤标志物CEA表达的清热解毒中药,指导临床应用。方法:用亚细胞毒浓度的药物作用于肠癌细胞一定时间后分别收集细胞培养液上清和细胞,分别采用夹心ELISA法和免疫组化法检测肠癌细胞培养上清CEA的表达。结果:所选取的清热解毒中药均有不同程度的抑制肠癌细胞生长的作用,并且随着中药浓度的递减,抑制率也相应的减低;夹心ELISA法检测培养上清夜CEA的表达在24h时,蚤休及黄连对CEA的浓度有明显的降低作用,有统计学意义(P<0.05);免疫组化检测CEA的表达显示:与对照组相比,黄连对CEA表达的阳性染色率最低,蛇莓、蚤休阳性染色率相当,夏枯草、连翘阳性染色率最强。结论:清热解毒中药对肠癌细胞株CEA的表达具有一定的抑制作用。
OBJECTIVE: To screen out heat-clearing and detoxicating traditional Chinese medicines that can significantly affect the expression of tumor markers CEA, and guide clinical application. Methods: Cells were incubated with sub-cytotoxic agents for a certain period of time. The supernatant and cell culture supernatants were collected. The CEA expression in the supernatant of colon cancer cells was detected by sandwich ELISA and immunohistochemistry, respectively. Results: The selected heat-clearing and detoxifying Chinese herbs all had the effects of inhibiting the growth of colon cancer cells in different degrees, and the inhibitory rate decreased correspondingly with the decreasing of the concentration of traditional Chinese medicine. The expression of CEA in the culture supernatant was detected by sandwich ELISA at 24 hours. The concentration of CEA was significantly decreased (P<0.05). The expression of CEA detected by immunohistochemistry showed that compared with the control group, the positive staining rate of CEA expression was lowest in the Coptis chinensis. The rate of positive staining of berries and pods was comparable, and the positive staining rate of Prunella vulgaris and Forsythia was the strongest. Conclusion: Qingrejiedu has certain inhibitory effect on the expression of CEA in colon cancer cell line.
