[目的]研究T-2毒素对人结肠癌细胞SW115的增殖抑制与凋亡作用。[方法]体外培养人结肠癌细胞SW115,加入浓度为1.6、8、40、200、1 000μg/m L的T-2毒素作用24 h,MTT法检测细胞增殖抑制率,流式细胞仪检测细胞凋亡,Hoechest 33258染色法观察细胞凋亡形态,并检测Caspase-3的活性变化。[结果]与对照组相比,T-2毒素为40μg/m L时显著抑制SW115细胞增殖(P<0.01),当T-2毒素达到200μg/m L,流式细胞仪检测凋亡率为27.55%,细胞内Caspase-3活性为0.597,差异具有统计学意义(P<0.01)。[结论]T-2毒素对人结肠癌细胞SW115具有明显的抑制作用,具有诱导SW115细胞凋亡的作用。
[Objective] To study the effects of T-2 toxin on proliferation and apoptosis of human colon cancer cell line SW115. [Method] The human colon cancer cell line SW115 was cultured in vitro. T-2 toxin was added at concentrations of 1.6, 8, 40, 200 and 1000 microg / mL for 24 h. MTT assay was used to detect the cell proliferation inhibition rate. Flow cytometry Apoptosis, Hoechst 33258 staining was used to observe the apoptotic morphology and to detect the changes of Caspase-3 activity. [Results] Compared with the control group, the T-2 toxin at 40 μg / mL significantly inhibited the proliferation of SW115 cells (P <0.01). When T-2 toxin reached 200 μg / mL, the apoptosis rate of SW115 cells was 27.55%, and the intracellular Caspase-3 activity was 0.597, the difference was statistically significant (P <0.01). [Conclusion] T-2 toxin has obvious inhibitory effect on human colon cancer cell line SW115 and induces the apoptosis of SW115 cells.
