目的:观察桔梗水提醇沉上清部分对α-葡萄糖苷酶活性的影响和对葡萄糖耐量缺损(IGT)小鼠糖耐量的影响。方法:先将桔梗进行水提,合并水提液置于65℃水浴浓缩至0.5g/ml浓度,用终浓度为50%的乙醇使其沉淀过夜,分离出上清于75℃水浴挥干乙醇制成流浸膏,使用前蒸馏水稀释到所需浓度。该有效部位在不同浓度下对于离体和在体的α-葡萄糖苷酶活性的抑制作用通过分光光度法测定;对STZ(一次性腹腔注射160mg/kg)造模IGT小鼠餐后血糖的影响通过3、6、12g/kg几个剂量的灌胃实验,并以拜唐苹作为阳性药,再测定餐后不同时段血糖浓度进行观察。结果:桔梗水提醇沉上清部分对离体和在体的α-葡萄糖苷酶活性均有显著的抑制作用,12g/kg的桔梗水提醇沉上清对IGT小鼠餐后各时段血糖的升高均有显著改善作用。结论:桔梗水提醇沉上清部分降低IGT小鼠餐后高血糖作用与其对α-葡萄糖苷酶的抑制作用相关。
