选取我国东北哈尔滨地区刺五加3个居群共49个单株的新鲜叶片,提取纯化基因组DNA。使用OPA和OPB系列8个随机引物进行PCR反应,对DNA样本做RAPD分析。3个居群多态RAPD位点的百分数分别为93.5%,91.3%和97.6%;3个居群内的平均遗传距离分别为0.241,0.363和0.388;3个居群两两单株之间的平均遗传距离分别为0.345,0.390和0.412。结果表明刺五加种内居群内和居群间的遗传多样性均较为丰富。为探讨刺五加种内变异及相关濒危植物遗传多样性分子机理研究提供了参考数据。
A total of 49 fresh leaves from three populations of Acanthopanax senticosus in Harbin, northeastern China were selected for purification of genomic DNA. Eight random primers from OPA and OPB series were used for PCR reaction and DNA samples were analyzed by RAPD. The percentage of polymorphic RAPD loci in the three populations were 93.5%, 91.3% and 97.6%, respectively. The average genetic distances among the three populations were 0.241, 0.363 and 0.388, respectively. The average genetic distance was 0.345, 0.390 and 0.412 respectively. The results showed that within the Acacia species within populations and populations genetic diversity are more abundant. This study provides reference data for the study on the molecular mechanism of intraspecific variation in Acanthopanax senticosus and related genetic diversity of endangered plants.
