目的 :检测人大隐静脉 (hum an saphenous vein,HSV)组织培养液中组织型纤溶酶原激活物 (tissue- typeplasm inogen activator,t- PA)的活性变化 ,及不同浓度纤维蛋白原 (fibrinogen,Fg)对 t- PA活性的影响 ,探讨血管移植后再狭窄发生发展的可能机制。方法 :应用 EAL AS酶标法检测组织培养液中的 t- PA活性变化。结果 :正常培养 HSV第 3、5天时的 t- PA活性水平均明显低于培养第 8、10、12和 14天的 t- PA水平 (P <0 .0 5 ,P <0 .0 1) ;低浓度的 Fg(0 .5 g/ L)对 t- PA活性无明显影响 ;高浓度的 Fg(2 .5 g/ L 或 5 g/ L)使培养 HSV第 3、5天时的 t- PA活性进一步降低 ,且降低培养 HSV第 8、10天时的 t- PA活性 ;但对培养 HSV第 12、14天时的 t- PA活性无明显影响。结论 :血管移植术后早期 ,移植静脉的低 t- PA活性水平与吻合口处的血栓形成相关 ;中、晚期的 t- PA活性升高则与血管平滑肌细胞的迁移及内膜增生等相关 ;较高浓度 Fg则具有促进这一过程的作用
OBJECTIVE: To detect the changes of tissue-typeplasm inogen activator (t-PA) activity in human humus saphenous vein (HSV) tissue culture media and the effect of different concentrations of fibrinogen Fg) on the activity of t-PA to investigate the possible mechanism of the development of restenosis after vascular grafting. Methods: The changes of t-PA activity in tissue culture fluid were detected by EAL AS enzyme-linked immunosorbent assay. Results: The t-PA activity of HSV cultured on day 3 and day 5 were significantly lower than that on day 8, 10, 12 and 14 (P <0.05, P <0.01) ; Low concentration of Fg (0. 5 g / L) had no significant effect on t-PA activity; high concentration of Fg (2.5 g / L or 5 g / L) PA activity was further reduced, and the t-PA activity of HSV on day 8 and 10 was decreased; however, the activity of t-PA on day 12 and 14 of culture HSV was not significantly affected. Conclusions: In the early stage after vascular transplantation, the level of low t-PA activity in the vein graft is related to the thrombosis at the anastomotic site. The increased t-PA activity in the middle and late stage is related to the migration and intimal hyperplasia of vascular smooth muscle cells. The higher concentration of Fg has the role of promoting this process
