目的 克隆人肺腺癌细胞单羧酸转运泵基因编码序列并对此序列作同源性分析。方法 通过RT PCR方法克隆人肺腺癌A5 49细胞单羧酸转运泵基因cDNA编码序列 ,应用PCR、四色荧光、双脱氧终止法测序 ;通过Genebank数据库应用分析软件对克隆基因序列进行同源性分析。结果 应用RT PCR方法克隆出人肺癌细胞的单羧酸转运泵基因 ;肺癌细胞此基因编码序列与人心肌细胞单羧酸转运泵基因第一亚型的cDNA编码序列具有高度同源性。结论 单羟酸转运泵基因可能为一保守的基因序列
Objective To clone the coding sequence of monocarboxylate transporter of human lung adenocarcinoma cells and to analyze the homology of this gene. Methods The cDNA coding sequence of monocarboxylate transporter of human lung adenocarcinoma A549 cells was cloned by RT PCR and sequenced by PCR, four-color fluorescence and dideoxy terminator method. The homology of cloned gene sequences was analyzed by Genebank database analysis. Results The monoclone transfer pump gene of human lung cancer cells was cloned by RT-PCR. The coding sequence of this gene in human lung cancer cells was highly homologous to the cDNA coding sequence of the first subtype of human carboxymethylcellulose transporter gene in human cardiomyocytes. Conclusions The monohydroxyacid transport pump gene may be a conserved gene sequence
