采用花粉管通道法,将青菜株系76F5、14号大白菜、紫阳等三个品种的总DNA及质粒pActl-DDNA导入青菜605品系,收获青菜种子.种子发芽后分别提取约两周苗龄的小苗总DNA,并经EcoRI酶切,以gus基因片段为探针做分子杂交.证实了外源gus基因已整合到青菜605品系中;田间观察还获得了一株与供体大白菜花形相似的变异株.
The total DNA and plasmid pActl-DDNA of Chinese cabbage line 76F5,14 Chinese cabbage and Ziyang were introduced into Chinese cabbage 605 line by using pollen tube channel method, and the vegetable seeds were harvested. Seed germination were extracted about two weeks seedlings young seedling total DNA, and by EcoRI digestion, gus gene fragment as a probe to do molecular hybridization. It was confirmed that the exogenous gus gene has been integrated into the strain of Chinese cabbage 605. In the field observation, a mutant strain similar to that of the donor cabbage was obtained.
