目的进行钩藤饮片生物碱组分(AUR)制备,探索AUR对D-半乳糖模型AD大鼠学习记忆功能作用。方法单因素筛选大孔吸附树脂制备钩藤饮片生物碱部位方法。采用D-半乳糖腹腔注射制备AD大鼠模型,采用Morris水迷宫测试大鼠的学习和记忆能力,采用分光光度法测定血清中乙酰胆碱转移酶(ChAT)、乙酰胆碱酯酶(Ach E)、乙酰胆碱(Ach)、超氧化物歧化酶(SOD)、丙二醛(MDA)活性。结果选择75%乙醇作为树脂洗脱溶媒。AUR能够增加AD模型大鼠的入水象限停留时间,缩短逃避潜伏期,提高血清ChAT、Ach、SOD水平,降低Ach E活性、MDA水平。结论 AB-8树脂适用于AUR制备,AD模型大鼠的学习记忆功能降低,AUR可改善AD大鼠的学习记忆能力,其机制与改善胆碱能系统作用有关。
Objective To investigate the effects of AUR on the learning and memory function of AD rats with D-galactose model. Methods Single factor screening macroporous resin preparation method for alkaloid parts of Uncaria. AD model was induced by intraperitoneal injection of D-galactose. Morris water maze test was used to evaluate the learning and memory ability of rats. Serum levels of acetylcholinesterase (ChAT), acetylcholinesterase (Ach E), acetylcholine Ach), superoxide dismutase (SOD), malondialdehyde (MDA) activity. Results 75% ethanol was chosen as the resin eluting solvent. AUR can increase the water intake quadrant dwell time, shorten the escape latency, increase the level of serum ChAT, Ach, SOD, decrease the AchE activity and MDA level in AD model rats. Conclusion AB-8 resin is suitable for the preparation of AUR. AD model rats have decreased learning and memory ability. AUR can improve the learning and memory abilities of AD rats. The mechanism is related to the improvement of cholinergic system.
