[目的]探明生防菌株枯草芽孢杆菌B-332的发酵条件及培养基配方。[方法]采用单因素和正交试验设计,通过摇瓶培养对枯草芽孢杆菌B-332菌株的发酵培养基和培养条件进行优化。[结果]最适培养接种时间为18h,优化后的培养基配方为:豆饼粉0.60g/L、蔗糖0.25g/L、硫酸铵0.07g/L、柠檬酸三钠0.03g/L、磷酸氢二钾0.003g/L、硫酸镁0.005g/L、硫酸亚铁0.0005g/L;发酵条件为:温度30℃、摇床转速180r/min、摇瓶装量80ml/500ml。在该综合条件下,发酵后的芽孢数为1.43×1011cfu/ml,比初始条件的芽孢总数4.03×109cfu/ml提高了34.48倍。[结论]该研究为枯草芽孢杆菌B-332菌株的工厂化打下了基础。
[Objective] The research aimed to investigate the fermentation conditions and culture medium of biocontrol strain Bacillus subtilis B-332. [Method] The fermentation medium and culture conditions of Bacillus subtilis B-332 strain were optimized by single factor and orthogonal design. [Result] The optimal culture inoculation time was 18h. The optimized medium formula was: soybean meal powder 0.60g / L, sucrose 0.25g / L, ammonium sulfate 0.07g / L, sodium citrate 0.03g / L, Dipotassium 0.003g / L, magnesium sulfate 0.005g / L, ferrous sulfate 0.0005g / L; fermentation conditions: temperature 30 ° C, shaker speed 180r / min, shake flask volume 80ml / 500ml. Under this comprehensive condition, the number of spores after fermentation was 1.43 × 1011cfu / ml, which was 34.48 times higher than the initial condition of 4.03 × 109cfu / ml spores. [Conclusion] This study laid the foundation for the industrialization of Bacillus subtilis B-332 strain.
