目的研究皮秒脉冲电场(ps PEF)对体外人脐静脉内皮细胞(HUVEC)生物学行为的影响。方法固定ps PEF脉宽800 ps、频率3 Hz、脉冲处理个数2000个,将HUVEC分为对照组和(200、300、400)KV/cm场强的低、中、高剂量处理组。在ps PEF处理HUVEC后2、4、6、8、10、12、14、16 h,CCK-8法检测ps PEF对HUVEC生长的影响,采用划痕试验及TranswellTM小室法观察ps PEF对HUVEC迁移率的影响,流式细胞术检测ps PEF对HUVEC细胞周期及凋亡的作用。结果随着场强的增加,细胞死亡率上升,生长受到抑制,12 h时抑制率最为显著;ps PEF处理可明显抑制HUVEC的迁移能力;ps PEF可将HUVEC阻滞于G2/M期,同时HUVEC凋亡细胞数增多。结论 ps PEF可抑制HUVEC增殖和迁移,并阻滞细胞周期于G2/M期并促进其凋亡。
Objective To study the effect of ps PEF on the biological behavior of human umbilical vein endothelial cells (HUVECs) in vitro. Methods The pulse width of ps PEF was fixed at 800 ps and the frequency was 3 Hz. The number of pulses was 2000. HUVECs were divided into control group and low, medium and high doses of (200, 300, 400) KV / cm field. The effects of ps PEF on the growth of HUVEC were detected by CCK-8 assay at 2,4,6,8,10,12,14,16 h after ps PEF treatment. The migration of HUVEC was observed by scratch test and TranswellTM cell method The effect of ps PEF on cell cycle and apoptosis in HUVECs was detected by flow cytometry. Results With the increase of field strength, the cell death rate increased and the growth was inhibited, and the inhibition rate was the highest at 12 h. The PS PEF treatment significantly inhibited the migration ability of HUVEC; ps PEF blocked HUVEC in G2 / M phase HUVEC apoptotic cells increased. Conclusion ps PEF can inhibit the proliferation and migration of HUVECs and block the cell cycle in G2 / M phase and promote its apoptosis.
