目的探讨异种〔猪/人)超急性排斥的发生机制。方法人血清为天然抗体和补体源。用四唑盐法行补体依赖的细胞毒反应,建立体外超急性排斥模型。结果正常人血清能溶解猪血管内皮细胞;而 Clq缺乏及B因子缺乏的人血清对猪血管内皮细胞的溶解率较低(P<0.01);同种猪血清及灭活补体的人血洁不溶解猪血管内皮细胞;将经典或旁路途径有缺陷的人血清等体积混合,其细胞毒作用恢复正常。结论治疗猪/人之间的超急性排斥应考虑补体旁路途径激活的问题。
Objective To investigate the mechanism of hyperacute rejection in pigs. Methods Human serum is a natural antibody and complement source. Using tetrazolium method to perform complement-dependent cytotoxicity, we established an in vitro hyperacute rejection model. Results Normal human serum could dissolve the pig blood vessel endothelial cells. However, the human blood serum lacking Clq deficiency and factor B deficiency had lower dissolution rate (P <0.01); the same pig serum and inactivated human blood serum were insoluble Pig vascular endothelial cells; the classical or bypass pathway defective human serum volume mixed, the cytotoxic effect returned to normal. Conclusion The hyperacute rejection in treatment of pigs / humans should consider the issue of activation of complement bypass pathway.
