目的:观察人趋化因子受体-4(CXCR4)重组腺病毒对胰腺癌SW1990细胞凋亡和细胞周期的影响。方法:用红色荧光蛋白重组腺病毒AdRFP和过表达CXCR4重组腺病毒AdCXCR4分别感染胰腺癌SW1990细胞做为阴性对照组和实验组,同时建立空白对照组,通过Hoechst33258染色和流式细胞术分别观察细胞凋亡和细胞周期的变化。结果:与空白对照组和阴性对照组相比,Hoechst 33258染色结果显示感染AdCXCR4能明显降低胰腺癌SW1990细胞的凋亡比率;流式细胞术结果显示感染AdCXCR4使SW1990细胞处于G_1期的细胞百分比明显减少,S期细胞百分比明显增多(P<0.05)。结论:过表达CXCR4可以抑制胰腺癌SW1990细胞的凋亡,促进其由G_1期向S期的转换。
Objective: To observe the effect of recombinant human CXCR4 on the apoptosis and cell cycle of pancreatic cancer cell line SW1990. METHODS: Pancreatic cancer SW1990 cells were infected with recombinant adenovirus AdRFP and CXCR4-overexpressing recombinant adenovirus respectively. The control group and blank control group were established. The cells were observed by Hoechst 33258 staining and flow cytometry Apoptosis and cell cycle changes. Results: Compared with the blank control group and the negative control group, Hoechst 33258 staining showed that the infection of AdCXCR4 could significantly reduce the apoptosis rate of SW1990 cells. The results of flow cytometry showed that the percentage of cells infected with AdCXCR4 in G_1 phase was significantly higher Decreased, the percentage of S phase cells increased significantly (P <0.05). Conclusion: Overexpression of CXCR4 can inhibit the apoptosis of pancreatic cancer cell line SW1990 and promote the transition from G 1 phase to S phase.
