目的:研究黄芪对晚期糖基化终末产物(AGEs)作用下人脐静脉内皮细胞(HUVEC)增殖和损伤的影响。方法:采用MTT比色法,观察黄芪对体外AGEs培养的人脐静脉内皮细胞增殖的影响;以内皮细胞膜与Alcain Blue(AB,阿利新蓝)结合量为指标,观察黄芪对AGEs导致的人脐静脉内皮细胞损伤的影响;利用AO/EB染料诱导AGEs作用下的人脐静脉内皮细胞,并利用荧光显微镜观察黄芪的抑制作用。结果:黄芪对人脐静脉内皮细胞增殖有抑制作用,可对抗AGEs导致的内皮细胞膜负电荷减少,并且对抗AGEs诱导凋亡的作用,对内皮细胞有保护作用。结论:黄芪是晚期糖基化终末产物的拮抗剂,可有效抑制人脐静脉内皮细胞增殖并有细胞保护作用。
Objective: To study the effects of astragalus on the proliferation and injury of human umbilical vein endothelial cells (HUVECs) under the influence of advanced glycation end products (AGEs). Methods: MTT colorimetric method was used to observe the effect of astragalus on the proliferation of human umbilical vein endothelial cells (AGEs) induced by AGEs in vitro. The binding amount of Alcian Blue (AB) The effect of AGEs on human umbilical vein endothelial cells was induced by AO / EB dye. The inhibitory effect of Astragalus membranaceus was observed by fluorescence microscopy. Results: Astragalus could inhibit the proliferation of human umbilical vein endothelial cells and reduce the negative charge of endothelial cells induced by AGEs, and could protect the endothelial cells against apoptosis induced by AGEs. Conclusion: Astragalus is an advanced glycation end-product antagonist, which can effectively inhibit human umbilical vein endothelial cell proliferation and cell protection.
