研究卡铂损伤灰鼠 (Chinchilla)耳蜗内毛细胞 (Innerhaircell,IHC)后 ,耳蜗核 (CochlearNucleus,CN)神经元出现的变化及可能涉及的环节。采取腹腔内给药 ,1月后取出用药组、对照组动物耳蜗 ,观察毛细胞 ,绘制耳蜗图 ,对耳蜗核以冠位连续切片 (10 μm) ,利用原位杂交的方法 ,以编码微管蛋白的cDNA片段为探针 ,进行检测。发现与耳蜗图所示全长内毛细胞破坏对应 ,伴随耳蜗核神经元体积缩小 ,耳蜗核内表达微管蛋白的细胞数减少。提示随着耳蜗内毛细胞的损伤、破坏 ,耳蜗核的传入神经冲动刺激被去除 ,影响了耳蜗核团细胞的信号系统的某些环节 ,从而导致细胞体积减少、细胞功能损伤等一系列变化。
To study the changes of neurons in Cochlear Nucleus (CN) and possible links involved in the carcinogenesis of Chinchilla inneris hair cells (IHC). After intraperitoneal administration, the cochleas were removed from the treated group and the control group after 1 month. The hair cells were observed and the cochlear map was drawn. The cochlear nucleus was serially sectioned with coronal slices (10 μm) Protein cDNA fragment as a probe for testing. Found that the cochlear map corresponding to the full length of hair cells destruction, accompanied by shrinkage of the cochlear nucleus neurons, the number of cells expressing tubulin in the cochlear nucleus decreased. It is suggested that with the damage and destruction of the hair cells in the cochlea, afferent nerve impulse stimulation of the cochlear nucleus is removed, which affects some aspects of the signaling system of the cochlear nucleus cells, resulting in a series of changes such as cell volume reduction and cell dysfunction .
