作琼脂对流免疫电泳时,一般需在琼脂板上打孔及去除孔内琼脂,操作不便。1978年应用预制打孔板作琼脂双向扩散试验。现经改进后,用作琼脂对流电冰,操作方便,简单易行,并探索了影响电泳的几个因素。由于降低了琼脂浓度,故琼脂胶透明度较好,微弱阳性亦可用肉眼观察到,例如 AFP 的最低可测量为400毫微克/毫升,兹介绍如下。一、材料与方法(一)制备模板取3毫米厚有机玻璃,裁锯长7厘米、宽3.5厘米作模板。以4毫米钻头打孔,孔距5毫米,行距4毫米。打12对孔。在模板一端两角及另一端中部以氯仿粘贴三个大小为3×2×2毫米的脚(见附图)。
