花药培养,是小麦育种的一种新技术。提高愈伤组织诱导频率和绿苗分化成活率,是花粉培养的关键技术问题。1980年我们应用合成培养基和简化培养基进行培养;用增产灵对接种前的穗子进行予处理;在诱导培养中,进行了照光和黑暗两种处理,明确了诱导效果。在绿苗移栽方面,初步明确了提高绿苗成活率的土壤条件。材料和方法供试材料为我所的小麦杂种一代,共21个组合。接种时的花粉发育阶段为单核中、晚期。接种前先用70%的酒精擦洗穗鞘,剥去穗鞘后,将各个组合分别扎成小捆浸入0.1%的升汞溶液中7—8分钟,用灭菌水冲2—3次,在无菌条件下接种,然后置于
Anther culture is a new technique of wheat breeding. Increasing callus induction frequency and green plantlet differentiation survival rate are the key technical problems of pollen culture.
