采用正交试验法研究深层发酵培养黑暗链霉菌F 2 11的培养基组成。实验结果表明 ,合理的培养基组成是 (% ) :葡萄糖 1.5 ;玉米粉 2 .0 ;硫酸镁 0 .5 ;玉米淀粉 2 .0 ;豆油 1.1;碳酸钙 0 .7;黄豆粉 3.5 ;硫酸锌 0 .0 1;硫酸铵 0 .7;淀粉酶 0 .0 2 5。实验结果还显示 ,在天然培养基中 ,无需再加铁、锰金属元素。研究发现 ,黑暗链霉菌孢子发芽对机械剪切敏感 ,利用孢子接种培养一级种子 ,采用空气搅拌 ,能得到良好的生长菌丝。菌丝浓度对暗霉素发酵生产水平影响显著 ,过高过低的菌丝浓度对菌体合成暗霉素均不利。采用改变空气流量来控制菌丝浓度 ,使其维持在推荐的曲线上 ,不至于过高或过低是一个好办法 ,可以有效地提高并稳定暗霉素的发酵水平
The orthogonal test was used to study the medium composition of Streptomyces coelicolor F 2 11. The experimental results show that the reasonable medium composition is as follows: glucose 1.5; corn flour 2.0; magnesium sulfate 0.5; corn starch 2.0; soybean oil 1.1; calcium carbonate 0.7; soybean flour 3.5; zinc sulfate 0 .0 1; ammonium sulfate 0 .7; amylase 0 .0 2 5. Experimental results also show that in natural medium, no need to add iron, manganese metal elements. The study found that the dark Streptomyces spore germination sensitive to mechanical shear, the use of spore inoculation culture seed, using air agitation, can get a good growth of mycelium. Mycelium concentration on the production of the level of production of significant impact on the production of alginic acid, hyphae hypothyroidism too much too low concentration of biosynthesis of phytoplankton are negative. It is a good idea to change the air flow to control the mycelial concentration so that it stays on the recommended curve, not too high or too low, which effectively increases and stabilizes the level of fermentation of the enzyme
