目的:研究白消安对雄性青春期小鼠生殖系统发育的影响。方法:108只雄性BALB/c小鼠随机分为对照组(n=24)和4个给药剂量的实验组(n=84)(10mg/kg 1次组,A组;10mg/kg 2次组,B组;20mg/kg组,C组;30mg/kg组,D组)。测量睾丸重量,HE染色观察睾丸组织形态学,并观察小鼠性行为和生育能力。结果:(1)组织形态学:第4周,各实验组的睾丸重量,B组的完全生精比例,B组、C组和D组的无生精比例与对照组相比有显著差异(P<0.05);第8周,C组的完全生精比例,D组的睾丸重量、完全生精比例和无生精比例与对照组相比有显著差异(P<0.05)。第12周,C组的睾丸重量,D组的睾丸重量、完全生精情况和无生精情况与对照组相比有显著差异(P<0.05)。(2)性行为学观察:C组和D组合并后与对照组相比,爬跨率和生育率存在显著性差异(P<0.05)。结论:白消安可损害睾丸组织发育,其毒性效应具有量效性和时效性。
Objective: To study the effect of busulfan on the reproductive system of male adolescent mice. Methods: One hundred and eight male BALB / c mice were randomly divided into control group (n = 24) and four experimental groups (n = 84) (10 mg / kg once a group, 10 mg / kg twice a day Group, group B; 20mg / kg group, group C; 30mg / kg group, group D). Testicular weight was measured. HE staining was used to observe the histomorphology of testes and the mice were observed for sexual behavior and fertility. Results: (1) Histomorphology: At the 4th week, the testicular weight of each experimental group, the complete spermatogenesis ratio of group B, the percentage of spermatogenic free of group B, C and D were significantly different from the control group (P <0.05). At the 8th week, the proportion of complete spermatogenesis in group C, the testicular weight in group D, the proportion of complete spermatogenesis and the percentage of spermatogenesis were significantly different from those in control group (P <0.05). At week 12, there was a significant difference (P <0.05) between testicular weight in group C, testicular weight in group D, complete spermatogenesis and no spermatogenesis. (2) Sexual behavior observation: There was a significant difference in creep rate and fertility rate between group C and group D (P <0.05) compared with control group. Conclusion: Busulfan can damage the development of testicular tissue, and its toxicity effect is dose-effect and time-effect.
