目的观察金黄色葡萄球菌肠毒素B(SEB)诱导脐静脉内皮细胞的凋亡效应。方法用不同浓度SEB感染脐静脉内皮细胞(HUVEC)2、4、8与12h后,用流式细胞术检测细胞凋亡率。结果SEB各浓度均可诱导HUVEC发生凋亡,与对照组比较差异有高度统计学意义(P<0.01)。当SEB浓度为5μg/ml时诱导细胞凋亡的能力最强;5μg/mlSEB作用HUVEC细胞2h后,即可诱导细胞产生明显的凋亡,且随着作用时间的延长,细胞凋亡率逐渐增加(P<0.01)。结论SEB可以诱导脐静脉内皮细胞凋亡,并表现出明显的时间、剂量效应关系,这可能与金葡菌L型垂直感染影响宫内胎儿发育的机制有关。
Objective To observe the apoptosis effect of Staphylococcus aureus enterotoxin B (SEB) on human umbilical vein endothelial cells. Methods Human umbilical vein endothelial cells (HUVECs) were infected with different concentrations of SEB for 2, 4, 8 and 12 hours. The apoptosis rate was detected by flow cytometry. Results The apoptosis of HUVECs was induced by different concentrations of SEB. There was a significant difference between the two groups (P <0.01). When SEB concentration was 5μg / ml, the ability of inducing apoptosis was the strongest; 5μg / ml SEB could induce obvious apoptosis of HUVEC cells after treated with 5μg / ml SEB for 2h, and the apoptosis rate gradually increased with the prolongation of action time (P <0.01). Conclusion SEB can induce apoptosis of human umbilical vein endothelial cells and show obvious time and dose-dependent effects. This may be related to the mechanism of L-type vertical infection of S. aureus affecting intrauterine fetal development.
