目的 EGFR19基因有19种常见突变型,临床检测分析一般很难分辨是何种突变型,探讨利用Excel软件制作EGFR19突变焦磷酸测序法参考图,提高工作效率。方法根据体细胞基因突变特点,假设EGFR19的突变型基因占20%,野生型基因占80%,将突变前后序列从2235号碱基起按顺序录入并按比例依次赋值,再利用Excel强大的图表生成和函数计算功能生成峰值模型图,经优化处理后可得到参考图。结果 EGFR19的19种参考图可根据录入的数据自动描点生成相应的参考图,实验室工作人员可灵活应用其对检测结果进行基因分型判断,进而更加合理的指导临床用药。结论应用Excel制作EGFR19的参考图具有操作简单、快捷准确和灵活性等优点,参考图有利于基因检测结果的分型。
Objective EGFR19 gene 19 kinds of common mutations, clinical testing analysis is generally difficult to distinguish what kind of mutation, to explore the use of Excel software to make EGFR19 mutation pyrosequencing reference map to improve work efficiency. Methods Based on the characteristics of somatic mutations, it is assumed that 20% of EGFR19 mutant genes and 80% of wild-type genes are EGFR19. The sequences before and after mutation are sequenced from 2235 bases in sequence and assigned proportionally. Generation and calculation of functions Generate a peak model map, the optimized map can be obtained. Results 19 kinds of reference pictures of EGFR19 can be automatically plotted according to the input data to generate corresponding reference pictures. The laboratory staff can flexibly apply the genotyping judgment to the detection results to guide the clinical medication more rationally. Conclusion The application of Excel to make the reference map of EGFR19 has the advantages of simple operation, quick and accurate and flexible. The reference map is helpful for the genotyping of gene test results.
