目的:观察青光安颗粒剂4种有效组分对兔眼滤过术后滤过道瘢痕组织TGF-β1和Smad3表达的影响,探讨青光安有效组分抗青光眼术后滤过道瘢痕化的效果及作用机制。方法:将青光安颗粒剂有效组分作用于滤过性手术后兔眼(有效组分1-4组),通过与正常组、模型组、丝裂霉素组(MMC组)的比较,观察青光安颗粒剂4种有效组分对青光眼术后滤过道瘢痕组织TGF-β1和Smad3的影响。结果:有效组分2组与MMC组TGF-β1和Smad3及m RNA表达比较差异无统计学意义,其余3组与MMC组TGF-β1和Smad3及m RNA表达比较,差异有显著性(P<0.05),其抑制效果不如MMC组。结论:青光安有效组分2和MMC都可通过抑制TGF-β1和Smad3 m RNA及其蛋白表达减少瘢痕组织增生,具有明显的抗青光眼术后滤过道瘢痕化的作用。

Results: There was no significant difference in the expression of TGF-β1, Smad3 and m RNA among the effective component 2 group and the MMC group, while there was significant difference between the other 3 groups and the MMC group (P < 0.05), its inhibitory effect is not as good as MMC group. CONCLUSION: SIK2 and MMC can both reduce scar tissue hyperplasia by inhibiting the expression of TGF-β1 and Smad3 m RNA and their proteins, and have a clear anti-scarring effect on glaucoma.
