目的观察异种(猪)脱细胞真皮基质(Xeno-ADM)对大鼠深Ⅱ度烧伤创面中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、白细胞介素-1(IL-1)表达的影响。方法取70只6~8周健康雄性Wister大鼠于背部建立深Ⅱ度烧伤模型,并随机分为治疗组和对照组,各组35只。治疗组大鼠烧伤创面采用Xeno-ADM覆盖包扎,对照组大鼠烧伤创面采用传统外用聚维酮碘换药包扎。按分组情况分别于伤后1、3、5、7、10、14、21 d每组各取5个大鼠,沿创缘取创面组织标本,采用免疫组织化学染色法和免疫印迹等技术检测创面愈合过程中炎症介质TNF-a、IL-8、IL-1的表达水平,采用SSPS 11.0统计软件对统计学数据进行处理,组间比较采用配对t检验。结果免疫组化提示:两组IL-8,TNF-a和IL-1均在第1天开始表达,第5天达高峰,第7~21天表达水平逐渐下降。其中,伤后治疗组的TNF-a、IL-8表达均低于对照组,且上述差异均有统计学意义(P<0.05)。而IL-1的表达在第5天到第7天实验组明显高于对照组,且差异均有统计学意义(P<0.05)。免疫印记显示,上述检测因子在两组中的表达规律及组间差异均与免疫组化结果相一致。结论 Xeno-ADM可有效抑制创面IL-8、TNF-a的表达和促进IL-1的表达,从而减轻炎症反应,促进创面愈合。
Objective To observe the effects of Xeno-ADM on the expression of tumor necrosis factor-α (TNF-α), interleukin-8 (IL-8), interleukin- 1 (IL-1) expression. Methods Seventy-two healthy male Wister rats aged 6 to 8 weeks were used to establish deep second-degree burn models on the back. The models were randomly divided into treatment group and control group, with 35 rats in each group. The burn wounds in the treatment group were covered with Xeno-ADM, and the wounds in the control group were wrapped with Povidone. According to the grouping conditions, 5 rats were randomly selected from each group on the 1st, 3rd, 5th, 7th, 10th, 14th and 21st day after injury. Wound tissue samples were taken along the margin and detected by immunohistochemical staining and Western blot The levels of TNF-a, IL-8 and IL-1 in inflammatory mediators during wound healing were calculated. Statistical data were processed using SPSS 11.0 statistical software. The paired t-test was used to compare the two groups. Results Immunohistochemistry showed that the expression of IL-8, TNF-a and IL-1 in both groups began to express on the first day, peaked on the fifth day, and gradually decreased from the seventh day to the 21st day. Among them, the expression of TNF-a and IL-8 in the post-injury treatment group was lower than that in the control group, and the differences were statistically significant (P <0.05). The expression of IL-1 was significantly higher in the experimental group than the control group on the 5th to 7th day (P <0.05). Western blot showed that the expression of the above detection factors in both groups and the differences between groups were consistent with the results of immunohistochemistry. Conclusion Xeno-ADM can effectively inhibit the expression of IL-8, TNF-a and IL-1 in the wound surface, and thus reduce the inflammatory reaction and promote wound healing.
