目的:探讨microRNAs(miRNAs)对原始卵泡的形成和发育的影响。方法:取1d龄新生小鼠卵巢,制备卵巢切片,利用原位杂交技术观察mir-143、let-7a、let-7b、let-7c和mir-125b在新生小鼠卵巢中的表达情况。结果:1d龄新生小鼠卵巢内只含有大量原始卵泡,mir-143、let-7a、let-7b、let-7c和mir-125b表达于这些原始卵泡的颗粒细胞内,在卵母细胞中不表达。结论:miRNAs可能通过调控原始卵泡颗粒细胞的增殖和/或分化,或通过调控下游靶基因表达直接或间接地影响卵泡形成和发育中的重要因子(FSH、抗苗勒氏管激素、生长分化因子9等),从而对原始卵泡的形成和生长启动产生影响。
