目的 建立动物耳鸣的行为学模型 ,确认动物是否感觉到了耳鸣 ,并用该模型评价药物治疗效果。方法 雄性健康Wistar大鼠 6 0只 ,平均分为 10组 :第Ⅰ、Ⅱ组为水杨酸组 ,皮下注射水杨酸钠每日 35 0mg kg体重 ;第Ⅲ、Ⅳ组为生理盐水对照组 ;第Ⅴ、Ⅵ组为纯音组 ,用 8kHz、6 5dBSPL的纯音代替水杨酸 ;第Ⅶ、Ⅷ组为尼莫地平组 ,注射水杨酸后再皮下注射尼莫地平每日 1mg kg体重 ;第Ⅸ、Ⅹ组为骨参舒耳片组 ,注射水杨酸后给予骨参舒耳片剂 5g kg体重。第Ⅰ、Ⅲ、Ⅴ、Ⅶ、Ⅸ组是从条件反射建立前开始给予药物或纯音 ,第Ⅱ、Ⅳ、Ⅵ、Ⅷ、Ⅹ组从条件反射建立后开始给予药物或纯音。将禁水后非常渴的动物置于隔声室内进行条件反射训练。持续背景白噪声强度 5 5dBSPL ,背景噪声随机停止为条件刺激 ,电击为非条件刺激。训练时给动物供水并记录吸水次数。经强化训练后形成“背景噪声停止 吸水减少或停止”的条件反射。不再给电击 ,观察条件反射的消除时间。结果 各组的消除时间分别为 :第Ⅰ组 5d ,第Ⅱ组 2d ,第Ⅲ、Ⅳ组均为 4d ,第Ⅴ组 5d、第Ⅵ组 2d ,第Ⅶ组 5d ,第Ⅷ组 4d ,第Ⅸ组 5d ,第Ⅹ组 4d。经双因素方差分析显示 ,各组之间和各训练时间 (d)之间差异具有极显著性意义 (P值均 <0 .0 0 1)。在此时间后动物吸
Objective To establish a behavioral model of tinnitus in animals and confirm whether the animal is feeling tinnitus and evaluate the effect of drug treatment by this model. Methods Sixty male Wistar rats were randomly divided into 10 groups: group Ⅰ and group Ⅱ were salicylic acid group, subcutaneous sodium salicylate 35 mg daily; group Ⅲ and Ⅳ were normal saline control group ; Ⅴ, Ⅵ group of pure tone group, 8kHz, 65dBSPL pure tone instead of salicylic acid; Ⅶ, Ⅷ group of nimodipine group, after injection of salicylic acid subcutaneous injection of nimodipine daily 1mg kg body weight; The Ⅸ, Ⅹ group Gushen Shu Otome group, after injection of salicylic acid Gushen Shu Er tablets 5g kg body weight. Groups Ⅰ, Ⅲ, Ⅴ, Ⅶ and Ⅸ were given drugs or pure tone just before the conditioned reflex was established. Groups Ⅱ, Ⅳ, Ⅵ, Ⅷ and Ⅹ were given drugs or pure tone after the conditioned reflex was established.
