目的为制定人鼠嵌合HCV感染小鼠模型做初步研究。方法将人骨髓干细胞,经肝脏对KM小鼠胎鼠,每只移植1×106人骨髓干细胞。用放射免疫法对移植小鼠肝脏中的甲胎蛋白进行免疫组织化学检测,分析人肝细胞在小鼠体内生长情况。结果移植后五周内可检测到甲胎蛋白。结论将人骨髓干细胞移植小鼠肝脏内能够存活并分化成肝细胞。
Objective To develop a mouse model of mouse chimeric HCV infection to do a preliminary study. METHODS: Human bone marrow stem cells and liver KM mice were transplanted with 1 × 106 human bone marrow stem cells each. Immunohistochemistry was used to detect the alpha-fetoprotein in the liver of the transplanted mice by radioimmunoassay. The growth of human hepatocytes in mice was analyzed. Results AFP was detectable within five weeks after transplantation. Conclusion The human bone marrow stem cell transplantation mouse liver can survive and differentiate into hepatocytes.
