目的:研究麻疯树核糖体失活蛋白的抗肿瘤作用。方法:抗肿瘤活性、N-糖苷酶活性和蛋白合成抑制活性分别用MTT法、苯胺裂解法和兔网织红细胞系统测定;用计算机软件分析比较麻疯树核糖体失活蛋白N-糖苷酶的活性结构域。结果:麻疯树核糖体失活蛋白具有抑制胃癌细胞(SGC-7901),小鼠骨髓瘤细胞(Sp2/0),人肝癌细胞体外增殖的能力,其IC_(50)值分别为0.23(0.15-0.32)mg/L,0.66(0.35-0.97)mg/L和3.16(2.74-3.58)mg/L,但对Hela细胞和正常细胞(MRC)无抑制作用。结论:麻疯树核糖体失活蛋白具有较强的抗肿瘤活性,其作用机制与其具有的N-糖苷酶活性相关。
Objective: To study the anti-tumor effect of Jatropha curcas ribosome inactivating protein. Methods: The antitumor activity, N-glycosidase activity and protein synthesis inhibitory activity were determined by MTT assay, aniline cleavage assay and rabbit reticulocyte system, respectively. The effects of N-glycosidase Active domain. Results: The Jatropha curcas ribosome inactivating protein could inhibit the proliferation of gastric cancer cells (SGC-7901), mouse myeloma cells (Sp2 / 0) and human hepatocellular carcinoma cells in vitro with IC 50 value of 0.23 (0.15 -0.32) mg / L, 0.66 (0.35-0.97) mg / L and 3.16 (2.74-3.58) mg / L, respectively, but had no inhibitory effect on Hela cells and normal cells (MRC). CONCLUSION: Jatropha curcas ribosome inactivating protein has strong anti-tumor activity, and its mechanism of action is related to its N-glycosidase activity.
