目的明确非甾体抗炎药(NSAIDs)对体外培养结肠癌细胞SW1116生长的影响及血管内皮生长因子(VEGF)表达的调控。方法采用MTT法观察NSAIDs对SW1116细胞生长的影响;电镜下观察尼美舒利诱导细胞凋亡的形态学变化;ELISA法检测细胞上清液中VEGF的含量。结果NSAIDs能呈剂量和时间依赖性抑制结肠癌SW1116细胞生长;加入尼美舒利后,上清液中VEGF的含量随作用时间的延长逐渐下降,作用72h后电镜下见凋亡小体。结论NSAIDs可明显抑制SW1116细胞生长,尼美舒利可诱导其凋亡,下调VEGF的表达,从而抑制结肠癌新生血管的形成。
Objective To investigate the effect of non-steroidal anti-inflammatory drugs (NSAIDs) on the growth of human colon cancer cell line SW1116 and the regulation of vascular endothelial growth factor (VEGF) expression. Methods MTT assay was used to observe the effect of NSAIDs on the growth of SW1116 cells. The morphological changes of the cells were observed by electron microscopy and the levels of VEGF in the supernatant were detected by ELISA. Results NSAIDs could inhibit the growth of SW1116 cells in a dose-and time-dependent manner. After adding nimesulide, the content of VEGF in the supernatant decreased gradually with the prolongation of time. After 72 hours, the apoptotic bodies were observed under electron microscope. Conclusion NSAIDs can significantly inhibit the growth of SW1116 cells. Nimesulide can induce apoptosis and down-regulate the expression of VEGF, thereby inhibiting the formation of colon cancer neovascularization.
