[目的]研究扶正养肝汤对免疫性肝损伤的保护作用及初步机制。[方法]60只昆明小鼠,随机分为6组,即空白组、模型组、联苯双酯滴丸组、扶正养肝汤低、中、高剂量组。建立刀豆蛋白诱导免疫性肝损伤动物模型,检测各组小鼠血清丙氨酸氨基转移酶(ALT)及天冬氨酸转氨酶(AST)水平;ELISA法检测各组小鼠血清细胞因子肿瘤坏死因子α(TNF-α)和γ-干扰素(IFN-γ)。[结果]扶正养肝汤低、中、高剂量组均能明显降低血清ALT及AST水平(P<0.01);细胞因子IFN-γ水平明显低于模型组(P<0.01);然而对炎症因子TNF-α无明显影响(P>0.05)。[结论]扶正养肝汤对免疫性肝损伤具有显著的保护作用,其机制可能与下调细胞因子IFN-γ表达有关。
[Objective] To study the protective effect of Fuzheng Yanggan Decoction on immune liver injury and its primary mechanism. [Methods] Sixty Kunming mice were randomly divided into six groups: blank group, model group, bifendate drip group and Fuzheng Yang Gan Tang low, medium and high dose groups. To establish a model animal model induced by concanavalin and to detect serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels in serum of mice in each group. Serum cytokine tumor necrosis Factor α (TNF-α) and γ-interferon (IFN-γ). [Results] The serum levels of ALT and AST in serum of the low, medium and high dose Fuzheng Yanggan Decoction group were significantly lower than those of the model group (P <0.01), while the levels of the inflammatory cytokines TNF-α had no significant effect (P> 0.05). [Conclusion] Fuzheng Yanggan Decoction has significant protective effect on immune liver injury, and its mechanism may be related to the down-regulation of the expression of IFN-γ.
