目的:研究加味温胆汤对大鼠营养性肥胖的影响,间接阐明中医“痰”与西医“脂肪”之间的相关性,为该方的临床应用提供可靠的实验依据。方法:采用营养性肥胖模型,将大鼠按体重随机分为7组,即空白组、模型组、千姿美组、曲美组和加味温胆汤高、中、低3个剂量组。除空白组外(普通饲料),其余各组动物均喂饲高脂饲料,连续5周,正常饮水。各组动物按不同的方法饲养5周后,将各高脂饲料组动物打乱重新分组,每组14只,雌雄各7只。分组后,口服给予相应的药物:千姿美0.22g·kg-1,曲美1.38mg·kg-1,加味温胆汤高、中、低3个剂量组分别为15.0、7.5、3.75g·kg-1,空白组和模型组分别给同体积的水,并记录各组动物每天的进食量,连续5周。各组动物末次给药后,检测血清TC、TG、LDL-C、HDL-C的含量;计算Lee’s指数、脂肪指数、全视野中的脂肪细胞个数,并用测微器测量脂肪细胞的大小。结果:加味温胆汤口服给药,可明显减轻高脂饲料引起的营养性肥胖模型大鼠的体重,降低血清TC、TG、LDL的含量,血清HDL含量明显升高,显著降低大鼠脂肪指数,增加全视野的脂肪细胞数量,缩小脂肪细胞体积。结论:该药具有对抗高脂饲料所致的营养性肥胖的发生。
Objective: To study the effect of Jiawei Wendan Decoction on nutritional obesity in rats, and to indirectly clarify the correlation between traditional Chinese medicine ”痰” and western medicine ”fat” to provide a reliable experimental basis for clinical application of this side. METHODS: Using a nutritional obesity model, rats were randomly divided into 7 groups according to their body weight, namely, blank group, model group, Qianzimei group, Qumei group and Jiawei Wendan decoction high, middle and low dose group. Except for the blank group (ordinary feed), the remaining groups of animals were all fed with high-fat diet for 5 consecutive weeks and normal drinking water. Each group of animals was reared for 5 weeks according to different methods. Each group of high-fat diet animals was disrupted and regrouped, with 14 animals in each group, 7 males and 7 females. After grouping, the corresponding drugs were given orally: Qianzimei 0.22g·kg-1, Qumei 1.38mg·kg-1, Jiawei Wendan Tang high, middle and low dose groups were 15.0, 7.5, and 3.75g respectively. Kg-1, blank group and model group were given the same volume of water, and the daily food intake of each group of animals was recorded for 5 consecutive weeks. After the last administration of each group of animals, serum TC, TG, LDL-C, and HDL-C levels were measured; Lee’s index, fat index, number of fat cells in the whole field were calculated, and the size of fat cells was measured with a micrometer. RESULTS: Oral administration of Jiawei Wendan decoction can significantly reduce the body weight of rats with nutritional obesity induced by high-fat diet, and reduce the content of serum TC, TG, and LDL. The serum HDL content increased significantly, and the index of rat fat was significantly decreased. Increase the number of fat cells in the entire field of view and reduce the volume of fat cells. Conclusion: The drug has an anti-fat diet due to the occurrence of nutritional obesity.
