探讨骨形成蛋白(BMP)对人骨肉瘤细胞的作用及意义.方法:将重组人BMPZ基因的反义核酸逆转录病毒表达载体PDOR-AB用脂质体导入BMP表达阳性的骨肉瘤细胞OS-99OI,观察骨肉瘤细胞OS-99OI的BMP表达、增殖活性、肿瘤抑制基因"m23的表达、细胞周期和电镜下形态改变.结果:转染后,OS-99o1细胞中内源性的BMP表达产物明显下降;增殖活性明显下降;肿瘤转移抑制基因"m23表达未见改变;细胞周期分析表明:GI期细胞比例上升,S期比例下降;电镜观察可见细胞质中溶酶体增多,染色质有断裂现象.结论:反义核酸技术阻断BMPZ在OS-99o1细胞中的表达后,肿瘤细胞的增殖活性降低,证实骨肉瘤细胞中BMP的表达增加了肿瘤细胞的增殖活性.
To investigate the effect of bone morphogenetic protein (BMP) on human osteosarcoma cells and its significance. Methods: The recombinant human BMPZ gene antisense nucleic acid retrovirus expression vector PDOR-AB was transfected into OS-99OI osteosarcoma cells with positive BMP expression by liposome. The expression of osteoclast osteoblasts OS-99OI in BMP, The expression of endogenous BMP in OS-99o1 cells was significantly decreased after transfection, and the proliferation activity was significantly decreased. The expression of “m23” was not found in the tumor metastasis suppressor gene The cell cycle analysis showed that the proportion of cells in GI phase increased and the proportion of S phase decreased. The lysosomes and chromatin in the cytoplasm were increased by electron microscopy. CONCLUSION: Anti-sense nucleic acid block BMPR expression in OS-99o1 cells, the proliferation of tumor cells decreased, confirmed that the expression of BMP in osteosarcoma cells increased the proliferation of tumor cells.
