目的:检测喉鳞癌中信号转导和转录激活因子3的活化和Skp2蛋白的表达,探讨其临床意义及相关性。方法:采用免疫组织化学法检测p-STAT3和Skp2蛋白在56例喉鳞状细胞癌和30例喉全(或近全)切除者癌旁(>2.0cm)正常黏膜中的表达,应用Metamorph/DP10/BX41显微图象分析系统测定阳性表达的平均光密度值,并进行统计分析。结果:与癌旁正常黏膜组相比较,p-STAT3和Skp2蛋白在喉鳞癌组织中过表达(P<0.01);p-STAT3和Skp2蛋白过表达与喉鳞癌的分化程度、临床分期、颈淋巴结转移有关(P<0.05或P<0.01);在喉鳞癌组织中p-STAT3蛋白和Skp2蛋白的表达呈正相关关系,相关系数r=0.8041(P<0.01)。结论:p-STAT3和Skp2蛋白过表达与喉鳞癌的发生、发展和转移等生物学行为有关,两者的表达呈正相关。
Objective: To detect the activation of signal transducers and activators of transcription 3 and the expression of Skp2 protein in laryngeal squamous cell carcinoma, and to explore the clinical significance and correlation. Methods: The expressions of p-STAT3 and Skp2 protein in 56 cases of laryngeal squamous cell carcinoma and 30 cases of laryngeal squamous cell carcinoma (> 2.0 cm) normal mucosa were detected by immunohistochemical method. Metamorph / DP10 / BX41 microscopic image analysis system to measure the average expression of the average optical density, and statistical analysis. Results: The expressions of p-STAT3 and Skp2 protein in laryngeal squamous cell carcinoma tissues were significantly higher than those in normal mucosa tissues (P <0.01). The over-expressions of p-STAT3 and Skp2 protein correlated with the differentiation degree, clinical stage, (P <0.05 or P <0.01). The expression of p-STAT3 protein and Skp2 protein in laryngeal squamous cell carcinoma was positively correlated with the correlation coefficient r = 0.8041 (P <0.01). Conclusion: The overexpression of p-STAT3 and Skp2 is correlated with the biological behavior of laryngeal squamous cell carcinoma, such as the occurrence, development and metastasis. The expression of p-STAT3 and Skp2 is positively correlated.
