目的观察高脂造模组大鼠肝细胞上B族Ⅰ型清道夫受体表达的变化,及辛伐他汀造模同时给药该受体表达的变化。方法雄性大鼠共60只,随机分为三组,每组20只,正常组以普通啮齿动物颗粒饲料饲喂,对照组以高脂组方与普通颗粒饲料饲喂,治疗组以上述高脂饲料饲喂,并同时以辛伐他汀(5 mg·kg-1·d-1)灌胃,共8周,采集血液标本,观察血脂变化;取肝脏制作冰冻切片,以辣根过氧化物酶标记的去载脂蛋白E的血浆高密度脂蛋白亚组分HDL3滴加到肝组织冰冻切片上,二氨基甲苯氨染色,普通光镜下观察,计算机图像系统分析。结果(1)与正常组相比,对照组胆固醇水平明显增高,组间差异有统计学意义(P<0.01);治疗组与对照组相比,胆固醇水平明显下降,组间差异有统计学意义(P<0.01)。(2)20倍光镜初步观察结果显示:正常组棕色沉积颗粒最少,对照组棕色沉积颗粒较多,治疗组与对照组相近。计算机图像分析系统分析可见:对照组阳性灰度值大于正常组(P<0.01),治疗组阳性灰度值接近于对照组。结论高脂血症时肝细胞B族Ⅰ型清道夫受体的表达明显增加,辛伐他汀降脂治疗不能增加其表达。
Objective To observe the changes of B-type Ⅰ scavenger receptor expression in rat hepatocytes induced by high-fat diet and the simultaneous expression of simvastatin and its receptor. Methods A total of 60 male rats were randomly divided into three groups of 20 rats. The normal rats were fed with normal rodent pellets and the control rats were fed with high fat diet and ordinary pellets. In the treatment group, The rats were fed with simvastatin (5 mg · kg-1 · d-1) for 8 weeks. The blood samples were collected to observe the changes of blood lipids. Frozen slices were taken from the liver and incubated with horseradish peroxidase Labeled apolipoprotein E plasma HDL sub-fraction HDL3 was added to the frozen sections of liver tissue, diaminotoluene ammonia staining, under ordinary light microscope, computer image system analysis. Results (1) Compared with the normal group, the cholesterol level in the control group was significantly increased, the difference between the two groups was statistically significant (P <0.01); the treatment group compared with the control group, the cholesterol level decreased significantly, the difference between the two groups was statistically significant (P <0.01). (2) The preliminary observation results of 20-fold light microscope showed that there were the least brown sediment particles in the normal group and the brown sedimentary particles in the control group, which were similar in the treatment group and the control group. Computer image analysis system analysis shows that: the positive gray value of the control group is greater than the normal group (P <0.01), the positive gray value of the treatment group close to the control group. Conclusions The expression of B-type Ⅰ scavenger receptor in hepatocytes is significantly increased in hyperlipidemic patients, and the simvastatin can not increase its expression.
