目的:建立测定中药络石藤中络石苷元-4’-O-β-龙胆二糖苷、络石苷、野漆树苷、络石苷元及木犀草素含量的HPLC方法。方法:75%甲醇提取络石藤药材,微孔滤膜(0.45μm)过滤,HPLC测定,采用Alltima C18色谱柱(4.6 mm×250mm,5μm),以0.5%HAc-甲醇为流动相梯度洗脱,流速1.0 mL.min-1,检测波长270 nm,采用外标法峰面积计算含量。结果:5种成分均达到基线分离,线性关系良好,精密度RSD<2.0%。该法成功的用于络石藤药材中5种成分含量的测定。药材中络石苷元-4’-O-β-龙胆二糖苷、络石苷、野漆树苷、络石苷元及木犀草素的平均回收率分别为98.27%,97.30%,97.20%,96.89%,96.48%。结论:方法操作简单、结果准确,具有较好的重复性和稳定性,为络石藤药材质量控制提供了依据。
OBJECTIVE: To establish an HPLC method for the determination of the contents of mesodextrin-4’-O-β-gentiobioside, treprostinil, palmatoside, trophosin and luteolin in the traditional Chinese medicine, Methods: 75% Methanol was extracted from Trachelospermi Caulis, filtered through a microporous membrane (0.45μm) and analyzed by HPLC. Alltima C18 column (4.6mm × 250mm, 5μm) was used to elute with 0.5% HAc- , The flow rate of 1.0 mL.min-1, detection wavelength of 270 nm, using external standard peak area calculation. Results: The five components all achieved baseline separation with good linearity and RSD <2.0%. The method was successfully used in the determination of five components of the content of Trachelospermi. The average recoveries of mesothelin-4’-O-β-gentiobioside, tretinoin, camphranin, treprostinil and luteolin were 98.27%, 97.30%, 97.20%, 96.89%, 96.48 %.
