目的探讨三氧化二砷(As_3O_3)对人胆囊癌GBC细胞化疗增敏作用及其机制。方法用流式细胞仪检测As_2O_3联合5-氟脲嘧啶、阿霉素和丝裂霉素作用下胆囊癌细胞凋亡情况。用Western blot分析凋亡相关蛋白Bcl-2、Caspase-3和PARP的表达。结果在As_2O_3的协同作用下,5-氟脲嘧啶、阿霉素和丝裂霉素对胆囊癌GBC细胞的诱导凋亡作用得到增强;蛋白水平的检测表明Caspase-3和PARP的剪切上升,而Bcl-2的蛋白表达下降。结论As_2O_3具备对胆囊癌细胞化疗增敏潜能,并主要通过改变Caspase-3、PARP及Bcl-2等的表达来实现。
Objective To investigate the sensitizing effect of arsenic trioxide (As_3O_3) on human gallbladder carcinoma GBC cells and its mechanism. Methods Flow cytometry was used to detect the apoptosis of gallbladder carcinoma cells treated with As_2O_3 combined with 5-fluorouracil, doxorubicin and mitomycin. Western blot was used to analyze the expression of Bcl-2, Caspase-3 and PARP. Results The apoptosis induced by 5-fluorouracil, doxorubicin and mitomycin on gallbladder carcinoma GBC cells was enhanced under the synergistic effect of As 2 O 3. The detection of protein level showed that the cleavage of Caspase-3 and PARP increased, The protein expression of Bcl-2 decreased. Conclusion As 2 O 3 possesses the potentiality of chemosensitivity to gallbladder cancer cells and is mainly achieved by changing the expression of Caspase-3, PARP and Bcl-2.
