目的:研究不同含锌饲料对幼鼠脑金属硫蛋白(Metalothionein,MT)表达的影响。方法:从哺乳期开始建立幼鼠低锌(ZincDeficiency,ZD)、常锌(ZincNormal,ZN)和高锌(ZincExcessive,ZE)动物模型,以流式细胞仪及免疫组化的方法,定位、定量观测脑组织(皮层、海马)内MT表达的变化。结果:幼鼠在哺乳期末(4周)和7周末脑组织内MT含量:ZD组明显低于ZN组,而ZE组明显高于ZN组。MT广泛分布于胶质细胞,散在分布于神经元内,在脉络膜、脑室管膜细胞及血管内皮细胞内含量丰富。结论:饲料锌明显影响幼鼠脑MT表达。MT可能对脑组织细胞内锌的代谢及生物学功能起重要调节作用,与脑发育有关。
Objective: To study the effect of different zinc-containing diets on the expression of metallothionein (MT) in young rats. Methods: Zinc Deficiency (ZD), Zinc (ZN) and Zinc (ZE) Zinc (Zinc) deficient animal models were established from the beginning of lactation. The animals were fixed by flow cytometry and immunohistochemistry. The changes of MT expression in brain tissue (cortex and hippocampus) were observed. Results: The contents of MT in young rats at the end of lactation (4 weeks) and 7th week were significantly lower in ZD group than those in ZN group and significantly higher in ZE group than those in ZN group. MT is widely distributed in glial cells, scattered in the neurons scattered in the choroid, ependymal cells and vascular endothelial cells rich in content. Conclusion: Dietary zinc significantly affects brain MT expression in young rats. MT may play an important regulatory role in the metabolism and biological function of zinc in brain cells, which is related to brain development.
