目的研究大鼠残肾模型肾脏组织学和缓激肽B2受体(BKB2R)mRNA表达水平的变化。方法24只雄性Wistar大鼠随机分成模型组和假手术组,每组12只。5/6肾切除术构建大鼠残肾模型。于造模前和造模后15、30、60、120 d分别检测大鼠尾动脉血压和血清肌酐水平。造模后1月和4月分别处死模型组和假手术组大鼠各6只,肾组织石蜡切片、PAS染色观察肾脏组织学改变,RT-real time PCR法检测肾组织BKB2R mRNA的表达水平。结果模型组大鼠造模后15 d血压和血清肌酐水平即较造模前明显升高(均P<0.01),以后逐渐上升至120 d达高峰;造模后1月残肾已出现肾小球代偿肥大,系膜中度增生,肾组织BKB2R mRNA的表达水平下调,4月时残肾出现明显的肾小球硬化、肾小管坏死和肾间质纤维化,肾组织BKB2R mRNA的表达水平进一步下调(P<0.01)。结论残肾模型大鼠在造模后15d即出现明显的肾功能不全,随后进行性加重,肾组织BKB2R mRNA的表达水平则逐渐下调。
Objective To investigate the changes of renal histology and expression of bradykinin B2 receptor (BKB2R) mRNA in rat remnant kidney model. Methods Twenty-four male Wistar rats were randomly divided into model group and sham operation group, 12 rats in each group. 5/6 nephrectomy to construct rat remnant kidney model. Rat tail artery blood pressure and serum creatinine were measured before modeling and 15, 30, 60 and 120 days after modeling. Six rabbits in model group and sham operation group were sacrificed in January and April after modeling respectively. The renal histological changes were observed by PAS staining. The expression of BKB2R mRNA in renal tissue was detected by RT-real time PCR. Results The blood pressure and serum creatinine were significantly increased at 15 d after modeling in model group (all P <0.01), then gradually increased to the peak at 120 d after modeling. Ball compensatory hypertrophy, mesangial hyperplasia, renal tissue BKB2R mRNA expression levels decreased in April when the residual kidney showed significant glomerular sclerosis, renal tubular necrosis and renal interstitial fibrosis, renal tissue BKB2R mRNA expression levels Further down (P <0.01). Conclusion The remnant kidney model rats showed obvious renal dysfunction 15 days after modeling, and then gradually increased. The expression of BKB2R mRNA in renal tissue decreased gradually.
