目的研究Gravin在血管内皮生长因子(VEGF)诱导人脐静脉内皮细胞(HUVECs)迁移和体外“小管样结构”(TLS)形成中的作用。方法用VEGF诱导HUVEC,通过Western blot和细胞免疫荧光分析Gravin在细胞内分布的变化;用G(o|¨)6983【蛋白激酶C(PKC)抑制剂】和VEGF共处理内皮细胞,观察对Gravin分布的影响,观察其对VEGF诱导HUVECs迁移和TLS形成的影响。结果VEGF呈时间和剂量依赖性诱导Gravin磷酸化,并向核周和细胞边缘重分布;以上效应能被0.5μmol/LG(o|¨)6983抑制;G(o|¨)6983同时抑制VEGF诱导HUVEC迁移和TLS形成。结论PKC-Gravin信号通路参与VEGF诱导的内皮细胞迁移和血管形成。
Objective To investigate the role of Gravin in the migration of human umbilical vein endothelial cells (HUVECs) induced by vascular endothelial growth factor (VEGF) and the formation of “tubular” TLS in vitro. Methods HUVECs were induced by VEGF, the changes of intracellular distribution of Gravin were analyzed by Western blot and immunofluorescence. Endothelial cells were co-treated with G (o | ¨) 6983 [protein kinase C (PKC) inhibitor] and VEGF, Distribution, and observe its effect on VEGF-induced HUVECs migration and TLS formation. Results VEGF could induce Gravin phosphorylation in a dose- and time-dependent manner and redistribute to the periphery of the perinuclear and nucleus. The above effects were inhibited by 0.5μmol / L (69.883) and G (o | ¨) 6983 inhibited the induction of VEGF HUVEC migration and TLS formation. Conclusion PKC-Gravin signaling pathway is involved in VEGF-induced endothelial cell migration and angiogenesis.
